You simply highlight the word and hit Alt+=. This opens the Add dialog box with your word and short form already filled in. Just hit Enter to accept the short form (or change it to one of your choice), and it's added to the current glossary.

Note that if you choose to make the short form more than one letter, Instant Text will ask you if you mean it and, if so, will put it in the phrase section.
